  • Tris is a buffer that's used to maintain a stable pH when working with solutions in the lab.
    In this video, we'll demonstrate the steps for preparing 250 mL of a 0.5 molar Tris buffer.
    We'll measure our powder, mix it with water using a stir plate, and then test our pH with a pH meter. We'll add drops of hydrochloric acid to lower the pH to our target of 7.8.

    Did you know that TRIS will form precipitate with Ag+ in conventional pH meter electrode?
    One does not simply measure TRIS buffer in pH meter with Ag/AgCl electrode with single junction reference electrode (mostly used in conventional pH meter now days) because it will poisons the reference electrode.
    using calomel electrode or double junction reference electrode is the correct way
    it seems that your using single junction electrode (i can see it clearly) and therefore your actual pH in not 7.8.

    I had no idea that you could add HCl to your buffer to alter the PH. my mind is blown.

    I have a question, if I want to prepare a solution at a concentration of 100 mM pH: 7.8 from this solution(0.5M, pH:7.8), I have to re-measure your pH? Thanks for you video.

    When i add the tris my solution becomes cloudy. I have to adjust the pH with hcl. Only after it will be clear again. I'm using 1 g/l of tris(THAM). Is this normal?

    good video ,,thanks alot,,, but can you help me about the best buffer that can be used in my experiment when my drug is unstable at phosphate buffer,,,, means that when i prepared the phosphate buffer the uv absorbance was not stable that changed with time but i need the same absorbance for 14 days !!!! so what the solution ?? plz help me :((

      You switched it. pH paper is not as accurate as pH meters

      joeriv5462 That's weird?!! In our biotech lab they prefer us using ph paper and said it was accurate. I think its because its faster...

      Hamburgo pH paper can be faster since all you have to do is insert it into your solution and look for color change. pH meters are quantitative so you can determine the exact pH of your solution.
