Thank you for this video! I have been trying to make some crystals from isopropanol/acetone (50/50 wt%), but I am finding it quite difficult to get the crystals to form. I am basing my procedure on: Horosanskaia, Elena, et al. "Purification of curcumin from ternary extract-similar mixtures of curcuminoids in a single crystallization step." Crystals 10.3 (2020): 206. However, my first step was to get the oleoresin directly by leeching into acetone and then filtering the solution, and then evaporating the acetone to get the resin. I am trying to grow the crystals using the dissolved oleoresin directly. So now I'm thinking that I might need to crystalize from the curcominoid mixture instead. After watching this, I think that perhaps the first step with hexane is used to remove the more non-polar components from the oleresin, and then with the acetone you get the curcuminoid mixture (demethoxycurcumin and demethoxy)curcumin and curcumin), which is then already purer and facilitates the crystallization. Is that correct?

The powder is prepared from the rhizome. Antimicrobial susceptibility can be measured for any solvent extract after drying by a rotary evaporator. Take accurately weighed dried extract (water, alcohol, ether, ethyl acetate etc.) and prepare stock solution like 1 ug/ml or 10 ug/ml 0r 100 ug/ml Then prepare different concentrations/dilutions of this stock solution like 0.2, 0.4, 0.6, 0.8, 1 ug/ml or 2, 4, 6, 8, 10 ug/ml or 20, 40, 60, 80, 100 ug/ml. Apply/add accurately measured dilutions to the disc by micropipette. Dry the plates, in a laminar flow hood for 30 minutes or more. Make sure the disks are fully dried before placing them on the plates. Once the disks are dry, flame-sterilize a pair of tweezers. Gently place the disk on top of the agar and lightly press it down with the tweezers. Incubate the plates as per protocol. sir how do i check the quality at home for curcumin or nano curcumin? can i check solubility at home ? if i purchased a nano curcumin - how do i check if it is nano curcumin or simple powder or extract ?
Nanosize of curcumin can be determined by a particle size analyzer (a sophisticated instrument). This instrument facility can be availed from chemical or pharmacy departments of state or central university or IIT institutes. Analysis cost may vary from 500 to 2000 INR per sample. To check solubility different solvents are required so it is always better to check solubility in nearby pharmacy colleges.
