I don't know in which river you collected your sample but 1600 cfu/100 mL sounds extremely high. I suspect that by multiplying the count on the 2 mL sample by 100 you are overestimating the amount of colonies on a hypothesized 100 ml sample, just because the count is likely not homogeneous across the whole water volume, so the 2 ml sample may not be representative of the count on a larger volume. Does that make sense?
if you want to keep something sterile, talking and breathing around it probably isn't a good idea, especially with plosives causing spittle everywhere.
No real need for gloves, unless you want make waste bigger for no reason. but do not put bulb pipettes on the desk, do not touch tip of the bulb pipette by the hands and mark Petri dishes on the bottom, not on the lids. And you will be good. ))
Why is it you emphasise not touching the sample bag apart from the tabs and then go ahead and touch the pipette before taking the sample with bare hands? I think your test is compromised.
