    She did not explain HOW the "factory" is updated, how was the underlying mechanism(s) of faulty NAD fixed or improved in order to make a better use of precursors such as NMN and NR.

      Hi, we have several blog posts on our website which go into further detail about how the ingredients in Nuchido TIME+ work to restore the cells ability to make and recycle NAD+. In summary Nuchido switches back on the NAD+ recycling system called the salvage pathway which is the main source of cellular NAD+ and it also inhibits inflammatory molecules CD38 which is known to increase with age and waste vast quantities of NAD+.

    She explains scientifically why NAD levels decline with age and how restoring them could have anti-ageing effects. This builds credibility.
    The data she presents on increases in NAD levels and related enzymes in people taking their product looks promising.
    Her analogy about supplying raw materials to an inefficient factory makes intuitive sense.
    The company appears to be using systematic drug discovery methods to identify ingredients. This is a plus.

    As she is the Lead Scientist for the company, she has an inherent bias to promote their product. More independent validation would carry more weight.
    The specifics of their "multi-target" formulation are lacking. What exactly is in it, and how do the ingredients work?
    While the short-term blood data looks good, long-term clinical trials on human health and ageing effects are needed.
    She provides little detail on the participants in the study - sample size, demographics, controls used, etc.
    There is no data on real-world customer experiences and satisfaction.

    Ha... That's exactly what I do. Multi systemic approach.
    I use L-tryptophan as it is part of the same pathway.
    I use luteolin, rutin and quercetin phytosome and apigenin from parsley for activation of CD38.
    I also add Inositol Hexanicotinate as it releases slowly, and doesn't cause a flush and I get the benefits of Inositol. I do also take som berberine TMG as they too work synergistically with the rest together with L-theanine.
    Been doing it for some months now and it feels better than just taking NMN.
    Next I will try adding in the version of NMN that Renue makes to see.kf I can boost the effect even further.

      I actually find nr to be better than nmn, but both are intermittently effective for me. Like 1/4 days and most days makes me feel crappy, even with tmg supplements, now I finally understand why.
      What’s that inositol you mentioned?
      Seems like I would benefit from something similar to your approach, can you help me understand it better? Which exact supplements are you using and where can I get them from?
      I already take Berberine & tmg.

    Absolutely brilliant wisdom from Dr. Conlon. Fixing the underlying problem of mito conversion of NAD+ makes so much sense. Plus, nicotinamide conversion is what the mito was designed to convert. Very timely important health information.

    Human aging is complex. I agree with multi-approach concept for anti-aging. Recently, Dr. Sinclair also use drugs cocktail for reverse cellular aging.

We have since conducted a larger randomised, double-blinded, placebo-controlled crossover clinical trial with Nuchido TIME+ the results of which are currently available on our website. This study is currently in the process of being submitted to a journal for publication, which we will share once it is available.

      Hi, yes absolutely optimising lifestyle habits are also important to increasing NAD+ levels. Exercise and fasting have both been shown to increase NAD+ and NAD+ production is linked to circadian rhythms so sleep is also very important.

      @@nuchido2094 yes, fasting and exercise can increase other functions like autophagy. Gut Biome also plays a crucial part. NAD is another important piece of the puzzle. It's surely better to have the whole body running more efficiently. NMN has helped, but I'm looking forward to my first shipment of Time :)

    A major reason is Older metabolism reactions create increased amounts of an enzyme CD38 which increaingly takes out NAD .. one needs to slow down production of CD38

    this talk is partly interesting information (NAD supplementation can be improved upon) but then avoids details of what to do about it for a marketing pitch for a proprietary blend of NAD with nutrients. To find the active ingredients you must visit the commercial portal offering $65 packs - to save you a click - the additions are from black pepper, green tea, quercitin etc. (from sophora japonica), R-ALA, vit-C. No particularly sensational breakthoughs there as far as I can tell - many sensible Perennials will already be taking these

    So far this looks good but i rather wait for longer term studies to see if results are maintained. Also we need to understand if the placebo group was very low, because the impact is high for 1 month. At these prices, better understanding is necessary

    Interesting video, thanks. We do have the Time+ product which we have tried based on earlier videos and information on the website. One question I have is that the amount of nicotinamide in the daily dosage (500mg) is 30x the RDA (in the US), which most people would classify as a 'megadose'. Since many people have become wary of megadosing in recent years, can you speak to the safety of such a high daily intake of nicotinamide?

    Looks good, can you explain how you address changes in methylation due to the increase in NAD+ levels or is that repaired as well? Also what about gout sufferers? We cannot supplement b3 and im assuming it’s relatives, or does the NAD+ level increases enable repair of the mechanisms involved in gout?

      Hi, Nuchido TIME+ does not cause methylation issues such as those associated with NR or NMN. This is because the breakdown product of NAD+ use called NAM is recycled back into fresh NAD+ as Nuchido switches back on the NAD+ recycling system called the Salvage Pathway. Therefore, the NAM is not accumulating and doesn't need to be methylated. NAD+ is known to activate cellular repair pathways, but we have not tested the product specifically in relation to gout.

      if you pour in percursors only, or let me say the mandatory b-vitamins, or even NAD+ into the serum, then you get side effects... quite strong, doing just the opposite, not boosting but draining. There is a physiological role of extracellular NAD , which gets extracellular if cells die, or die soon, eg. if they got hijacked by a virus. similar with high concentrations of precursors.
      If you fix the process, the story is a whole lot different. Of course, I would recommend not to boost to levels of a baby, or even pre-pubertarian. These young organisms are very different. But restoring levels as to look like from a 25y old body, I would not expect any side effect.
      That being said, there is a risk for awakening cancer cells that a body has already required, like decades ago, when in your youth you have been heavily smoking or the like. Such, the treatment should be slowly ramping up, supported by anti-cancer food, and clearing senescent cells.

While the boundaries between them are fluid TRF means to eat only ina certain time window of 4..8 hours., resulting in a non-eating window of 16h +/-2h . Some say it is preferable to eat early, while for some others that may not be possible due to external settings.

      ATP is the energy created by the Mitochodria which relies on NAD to do this. All cells need NAD including cancer cells, which is why there has been doubt about it's use by people with cancer. However....NMN helps prevent cancer and has been shown to halt or send certain cancers into remission. The only time it has been shown to aggrevate cancer is when an aggressive cancer has been induced in mice. When you consider that poorly functioning mitochodria are the most major cause of cancer and that nad precursors help repeair mitochondri, you are actually tackling cancer at it's source. Also consider that all foods can help promote the growth of cancer. Weigh it up....without NAD...you're dead....with NAD you have a greater chance of fighting cancer.

      Hi, there is no evidence that NAD+ causes cancer. What experiments do show is that NAD+ production can be targeted as an Achilles heel in cancer cells. If you cut off the NAD+ supply cancer cells die. The same applies to glucose and oxygen. Cancer cells are heavily dependent on a supply of critical molecules and removing the supply locally kills cancer cells, but this does not mean glucose, oxygen or NAD+ cause cancer, nor is it a sensible approach to restrict any of these critical molecules. These experimental observations are simply one of many alterations in a cancer cell that evolve to confer a survival advantage and as a result researchers are quite rightly pursuing this avenue as a potential cancer therapy.
      NAD+ levels and cancer risk are actually strongly negatively correlated throughout life. Cancer risk rises as NAD+ levels decrease with age, both exponentially.

      @@dwdwone Simple answer: Increased NAD levels enhance glycolysis and fuel cancer cells. And....Cancer growth is predicted to require substantial rates of substrate catabolism and ATP turnover to drive unrestricted biosynthesis and cell growth.
      And....emerging evidence suggests that cancer is a mitochondrial metabolic disease.
      It follows then that looking after the mitochondria helps prevent cancer.
      NAD precrusors have been shown to halt or reverse certain types of cancer but have also been shown to aggravate cancer in mice who have purposely been given an aggressive form of cancer.
      NMN helps prevent cancer but accellerates aggressive cancer, so as I indicated before it may become a race between repair or worsen. Anyone with cancer thinking about taking a NAD precursor would be well advised to take advice from a a doctor, especially one who is familliar with such pre-crusors.

      But you also feed the immune system, which fights cancer.
      I guess that it's positive if you don't already have cancer.
      But negative if you are trying to starve cancer, as you do with typical cancer therapy.

      You shouldn't expect NAD in the ingredients, because that large molecule don't go into the cells.
      That's why you use the precursors, as the video says.
      They utilize NAM as precursor, which relies on the salvage pathway. And a few ingredients which are intended to reduce the consumption of NAD.
      I don't know if their product works, because I don’t see why it would improve the salvage pathway, though.

      Hi, as the other comment states NAD+ itself is a large molecule, which struggles to enter cells. That is why cells have in built NAD+ production pathways like the salvage pathway, which is the main source of cellular NAD+. Our formulation contains the precursors NAM which is the natural breakdown product of NAD+ use by the cell and is therefore the body's preferred precursor as it can easily enter cells without a transporter. Our formulation contains ingredients to switch back on the NAD+ recycling systems and inhibit waste of NAD+ via inflammatory molecules like CD38. We have conducted a clinical trial on our product which showed an increase in NAD+ within just 7 days. A reduction in inflammation and glycation., activation of NAMPT (the enzyme that powers the salvage pathway), increase in sirtuins and a reduction in biological age after just 28 days.     There was 10 minutes of explaining why precursors alone are not enough, but no explanation of what the "multi-pathway" approach is actually doing, what is in it, or how it works. I'm sure there is legitimacy but this is a sales video, not helpful information.

      Apigenin to inhibit cd38 enzyme and 5 amino 1mq to help the nmnt pathway. There may be better ways to do it but those are places to start

      there no explanation in this one, but there are others, just search for her, not only a sales video, the whole channel is sponsored by a few start up companies. they use NAM, a plant compound, which helps the rate limiting enzyme of the salvage pathway, and apigenin == 3 targets here. + some trivials

    There are many molecules that decline with age. All of the simi essential amino acids and non essential amino acids decline with age. Supplementing helps but doesn’t cure the problem.

      Hi, yes niacinamide/ nicotinamide (NAM) are both precursors used by the cell to make NAD+. NAM is actually the natural breakdown product of NAD+ being used by the cell and is included in the Nuchido TIME+ formulation.

      Hi, there are various feedback loops within cells but the salvage pathway recycles NAM back into fresh NAD+.

    What about the Brexit. Do you have storage facility within the EU to ship from? So that the customer may have a good customs experience? Are you going to lower the price over time as sales go up, or is this price staying? Label mentions quite a few cheaper raw materials. Like the quercetin source seems to be the cheeper raw material out there. How do we know this is not another snake oil? I don't mean to say that more affordable raw materials is a negative thing. But then you can find a middle ground for profit within time as well?

Conducting this level of testing is very time consuming and costly.     Why decline in enzyme? What’s the natural solution? Prescription of drug does not make sense as you don’t know fixing this will damage another factory line. Half knowledge is more dangerous than no knowledge…Best bet focus on lifestyle n nutrition
