Shouldn't his spider sense have warned him of the danger of that handshake? Or is that not working anymore?
usm still didn't understand how the Spidey sense worked at this point since this was his first/second month on the job.
What I want to know is why didn't his spider-sense go off?
It canonically doesn’t warn Spider-Man against his own stupidity.

This is a less experienced spiderman, also his spider sense is canonically just a 4th wall break but peter can't tell the difference between that and his subconscious, so he chalks it up to a bad feeling, this means he'd probably think it's a trap when going to shake shockers hand, and might even warn himself to move out the way, but a inexperienced spiderman might just see that as them being paranoid.

@booshandorkakow64 You could say the same for every Spiderman, but they've all been jumped or tricked before.

What's the context for this?
This is from the new Ultimate series by Jonathan Hickman, and this peter parker was bitten by a radioactive spider during his adult years instead of his teenage years, and he's trying to figure out how to be spider-man after learning that he was supposed to BE spider-man during his younger years.
In "Ultimate Invasion" (2023), The Maker (evil version of Reed Richards) breaks out of prison. He steals several items from multiple nations (not because he necessarily needs them but to mock the heroes that he can do so without being caught in time) and creates a machinery, that is letting him jump through the multi-verse (I forgot which one). There he either prevents "canon-events" such as Peter getting bitten by the spider in his younger years, delaying the rocket launch from Fantastic Four, which prevents them from getting hit by the nova which gave them their powers, et cetera... some heroes he even killed. Some years down the drain, heroes from the future (variants from Cap, Thor, Wasp) invade him (The Maker), but they fail. They do manage to kill Obediah Stane and knock out Howard Stark (Iron Man in this universe). Howard recovers and the maker tells him about his scheme (which he also forgot partially because someone blasted parts of his head away). He wants Howard to build a replica of the Immortus Engine, with the help of a captive Reed Richards (who wears a mask similar to Dr. Doom's). Howard has other plans and builts something else. He shows it to the Maker (distracting him, while Richards/Doom steals stuff from the Maker, giving him the instruction to entrust those things to Tony Stark), when suddenly a variant of Kang arrives with yet another batch of variants from Cap, Thor and the Wasp. The Maker remembers, that he's the same person who blasted off parts of his head. They fight and Howard dies. Richards/Doom, as instructed, gives Tony the items (that also includes the recordings of the heroes from the other universes) which he stole from the Maker, they build a Suit and Tony dons himself Iron Lad instead of Iron Man, because he's not yet earned the title of his late father. He records hologram messages to multiple people, who were supposed to become heroes (like in the other universes), leading to the events of "Ultimate Spider-Man" (2023-), "Ultimate Black Panther" (2024-), et cetera...
At the end of the first issue from Ultimate Spider-Man, Peter receives a box with said message, and a vial with the spider, that was supposed to bite him 20 years ago.
