Hi there plants in jars , love your videos , I’m thinking of buying a tissue culture kit just to do this as a hobby, love to do mushroom culture in Petrie dishes to and develope the spores to make more , tried it once and failed lol wasn’t sterile enough , lessens to learn hey . I do think I can do this , very fascinating . Tissue culture plants will be loads of fun , I get a buzz out of things like this. 👍
You absolutely have to do an episode with Dickson Despommier and Vincent Racaniello from TWIV, yes I know it is a Virology podcast but the platform those two built , with new shows like Matters Microbial they intersect with yours in major ways and such giant resources for the people who love staying up on discovery week by week, paper by paper. i know everyone has a precious list of advice but I sorta think you could expand your viewers and I know Vincent is all about it.
I have had luck using that media for carnivorous plants!
3:17 what do you think about sterilizing the plants with e.g. 6% H2O2 instead of bleach?
could we also put the plant material cleaned with H2O2 directly into our culture medium without cleaning with disinfected water first?
I have seen in vitro plants that were in water with prmanent 2% H2O2 and they looked great.

Not hating or anything but is there a reason why chose the orchid multiplication medium as opposed to the orchid stem propagation medium?
Hey nice video! I have a similar lab setup for mushroom cultivation and wanted to get back into plant tissue culture and so glad I found your channel, I appreciate the detailed video! My advice from past experience is maybe the initial cuts were not as sheer/clean as possible with those scissors - try using a sterile scalpel blade in a petri dish while using the forceps - it could help with the contams and tissue die off by making a less damaging cut
I don’t use a scalpel :) if you watch some of my other videos you’ll see why lol

Do we have to use any chemical to break the bud dormancy in MS media?? My Dendrobium bud got stuck after some time and it's not developing

There is a book called Plants from Test Tubes that is really helpful. It's not specific to orchids but it does have lots of good information about tissue culture!

how do you keep track of the orientation of your explants through the process?
If it's a plant where I can't tell just by looking at the explant then I'll keep one of the internodes longer to remember

When you put the jars in the autoclave, are the lids just resting ontop of the jars as a cover or are they screwed on tight?

Tried different methods and media.
Contaminations or burning the explants when not contaminated.
When non contaminated, no germination and no callus formation.

Could you pls make a tutorial about growing ghost orchid with tissue culture? I was wandering when I saw the video about the Venus flytrap propagation that these orchid is also in great demand an great risk of extinction, they don't need air movement so maybe these would be a good solution

Idk if I will ever make a specific tutorial but I am currently growing ghost orchids :) Unfortunately the main supplier of them in Florida is an a$$
You avoided BAP on the other video I watched of yours, but this has it. Is it just sorta mostly dangerous unless you're careful?
If you wear gloves and avoid ingesting it / getting it in your eyes, then it's really fine to use. I knew a lot of people would be trying TC for the first time from my other video, so in that case I always recommend starting with Kinetin. Easier to dispose of, etc.
@@plantsinjars But keep in mind it's a Class 1 Carcinogen that CAN pass through skin. The Kinetin advice is genuinely very good.
How do I dispose of the culture media after I deflask? Can I pour it down the sink?
I scoop it into a trash bag and throw it away. it only contains trace amounts of PGR’s

I can’t even keep an orchid alive. I’m jealous.
I used the following explant sterilization for J.regia (persian walnut tree) --> 30sec in ethanol 60% solution then washed 3 times in sterile tap water then 30min in a mixture of captan and benomyl fungicide (each 1gr/L) + 2mg/L streptomycin (antibiotic) and then washed 3 times and then 15min in a bleach solution and then again washed 3 times with the same water. -------------⚠️
but no matter how hard I try, my explants dies after a few days due to endogenous contamination. What can I do to prevent this? I even added 50mg/L streptomycin in my establishment phase medium but still no luck 😭
-ps: I use nodal segments of a walnut tree in the field as my initial explants

Hi- is it possible for me to do meristem tissue culture at home? I’ve watched your tissue culture videos and I think I’m totally capable of tissue culture. I grow dahlias and I had to toss more than half my dahlias (I grow about 500, many rare) this year due to virus. One is my own breed that is named after a dear relative who passed away. I think it may be diseased and I am heartbroken. I think meristem culture is the only way to eliminate the disease.
With a microscope and lots of practice it is definitely possible
@@plantsinjarsany videos you can point me to? From drawings, I can quite see where on a plant I would find the meristem.
Also, what low cost microscope would you recommend?

They grew shoots but then got contaminated. I needed to seal the jars with Saran Wrap

I dont trust my PH meter either. Those things are diabolical.

I HATE MY PH METER SO MUCH TOO. My calibration solutions never read accurately and it seriously pisses me off. If you ever find one that works right and doesn't keep people up at night please make a video about it.
The bluewave one is decent as long as you store it in kcl solution but it's so overpriced.
