Achara is also right when she says ratri is night as it's from a Sanskrit origin.
samay samapti (time end) is basically the announcement made by Amitabh Bachhan in KBC (who will be a millionaire) when the contestant's time runs out.
FYI 3:56 is how Amitab Bacchan ends Kaun banega crorepatti ( Indian who wants to be a millionaire). the dialoue is Samay Samapti ki ghoshna (we have no more time left).

Achara is correct, Samay is literally time. Ratri means night in Hindi and Sanskrit. While Raat is night in "common use" Hindi. Also the AmitabhBachchan angle is that he hosts "Kaun banega Crorepati?" the Indian version of "Who wants to be a millionaire?". So in the show it is him who tells the contestants, your "time" starts now or aapka "samay" shuru hota hai.. ab. That's what he's referring to.
Amitabh also says "Shubh-ratri" at the end of KBC which literally translates to "Good Night"
In Telugu we say "ratri" as well.
