Shes been retconned a bit. She was a basically space demon who took the form of a twin in the womb with Xavier with the intention of replacing him. Baby Xavier killed her in vitro. Then, she came back still evil but actually believed herself to be his sister and wanted revenge. She blew up Genosha and killed millions of mutants. She came back a few more times until Jean Grey forced the empathy of the world into her mind which just made her evil for mutants (instead of against them as she was before). Yes, I wrote all of that.

Her big story line is that in the comics she causes the story line that happens in Episode 5 of X-men 97.

She killed millions of mutants in Genosha and what episode 5 of Xmen 97 is based on.

It looks like it's a mixed of both Xmen 97 episode and Old Man Logan. You can see Pym Falls in the scene. Which means not just Xmen, but 99% of Marvel super heroes got killed in Cassandra's universe.

Oh, they definitely showed some of the cameos, they just weren't on screen for long enough for you to really get a good look. But if you pause at the right time, you can clearly see who all might be coming back, and I am SO FREAKING EXCITED for some of them!

Probably already beaten to it by other fellow Peaches, but just in case -- the bald woman is Cassandra Nova, Charles Xavier's twin sister, introduced during Grant Morrison's run of X-Men comics. She's so evil that Charles "killed" her while they were still in the womb but she survived.
They actually did show cameos, but it was really quick. When the Giant Man helmet opens, you can see Lady Deathstrike and Azazel (from First Class).

Hugh's smile when he leaned into Deadpool's gun at the start was a weird combination of funny and depressing.

So psyched!!!!! The most I’ve been, in awhile, for a movie.
Cassandra Nova is what's known as a Mummudrai or an "anti self" that was born without form on the astral plane. She latched onto Charles in the womb and became his dark counterpart, taking on all his abilities. When she had formed hands she tried to strangle him with his own umbilical cord. He tried to destroy her with a psionic blast which caused her body to be stillborn, only existing as chaotic cellular matter. She'd spend several decades in the sewer rebuilding human form and learning how to blend in as human. Then she went to work destroying everything i think that about sums it up.

Uh, yeah ... I wouldn't get my hopes up. Not after the nonsense we saw in She-Hulk. As a longtime Daredevil fan, gong back to the early 70s, I was happy to hear that Disney ended up scrapping something like eight episodes they had shot of the new series, and were starting over. I can only assume that even lame, woke-as-hell, let's-girl-boss-every-damn-thing Disney realized that Daredevil can't be screwed with that way. Then again: it's Disney; if nothing else, they've shown that they'll screw things up for that sweet DEI influx of money from Vanguard, Blackrock, etc. That's probably how we got the lame-as-hell female Silver Surfer ... and why another few thousand people recently canceled their D+ accounts. Face it: the MCU is basically dead at this point. Ryan Reynolds is single-handedly keeping the corpse animated with some fun zombie-dancing, but that's about it. Unless Disney changes their tack, and actually starts respecting their audience, they're going to continue to lose integrity, fans, and money.

Bernthal is back as Punisher, which he said he absolutely wouldn't do unless it went R-rated, they have the same writing team from the Netflix series, Karen and Foggy are back, D'onofrio and Cox both say the scripts are amazing, we know there's going to be blood-how much more is it gonna take for people to accept that the MCU might just possibly be swinging back into the good zone after a string of screw-ups? Ye of extremely little faith.

I think in the shot where Giant-Man’s dead head opens, we can see the guy who Wolverine kicked the balls of in X-Men: The Last Stand, the one who he told to “grow those back.”

According to the Shi'ar, everyone fights their evil side before they are born, which is basically the mummudrai like Cassandra, so if the person loses, the mummudrai is born.

After someone mentioned in the comments on another video that the big bald baddie is Cassandra Nova, I did some Googling (since I'm not much of a comicsman, more of a movie/TV guy). The story I've pieced together: Cassandra Nova is Xavier's evil twin sister and also astral anti-Charles according to the Shi'ar. In the womb, Charles could sense that Cassandra was evil, so he killed her, causing a miscarriage. But the clump of cells that would be Cassandra survived in the sewer (because comics, I guess... comics are weird) and developed into a full person. She's got basically all of Xavier's powers, but seeks revenge on him and his X-Men because he killed her.
...yeah, I'll say it again: comics are weird, man.

They did show some cameo xmen cameos in the larger shot of Ant Man Skull. Its gonna be dope!!! And of course everyone is mentioning the big bad as Cassandra Nova which is a storyline of comics I never read but sounds ridiculous. I am here for it!

I love how meta the marketing for this has been. They first announced it in such a tongue in cheek way and this trailer just ran through everyones pipe dream expectations in an efficient manner while also hinting at more.

This is going to be completely unsupported by any evidence and motivated purely by my own dreams, but I'm calling it - that magic portal at the end of the trailer, leads Deadpool and Wolverine straight to the battle from Endgame. We need them "properly" in the MCU and this could be the connection.
Everyone is pointing out that bald lady is Cassandra Nova, but what I haven’t seen is anyone point out that Cassandra Nova broke all our hearts (sort of) 2 weeks ago.
In the comics, Cassandra Nova was responsible for the Wild Sentinel attack on Genosha that killed 16.5 million people.
