Enzymatic inhibition by drugs, Part 1

Why is it that irreversible enzymatic inhibitors like aspirin and NSAIDs affect platelets for the life of the platelet (9 days) but only decrease inflammation for four hours? How can our drugs affect enzymes? So much to learn about enzymes and how our medications affect them but learning about it is interesting.
