Ryoba's laugh sounds a lot more natural than ayano's, the thing is that she really sounds like she founds funny what she's doing meanwhile ayano laughs when she needs to calm down, not because she founds great or hilarious what she's doing unlike ryoba.

Well according to the lore ryoba is more "human" than her own daugther which mean she still feels stuff and thing, that's why her laugh sounds more like she is more real and more "human" as she feels fun while doing her stuff. While ayano is just doing that to keep her sanity up, she is not doing her thing for fun but she's doing her things because she had to. I think that makes Ryoba is scarier and more dangerous.

You can tell that it isn’t as forced as Ayano’s…Ayano’s laugh sounds like she’s almost making herself out to be evil and that she is just laughing at the overwhelming loss of sanity and feelings that she gets.
