    You could have made a 4 quadrant streak to isolate specific colonies, then do various simple stain techniques (gram stain) to view it under a microscope for easy determination of what you have on each plate. I remember in microbiology we tried the sanitizer and still found some bacteria because when you use the sanitizer there is ALWAYS natural bacteria coming out of our skin which every human is born with. I can imagine how crazy someone can get after realizing how much bacteria is everywhere.

    Not at all. All it means is that there is a greater variety of species on the keyboard than in the bathroom. Remember, the nutrient agar is creating an optimal environment for this bacteria to grow. It isn't an accurate representation of how much bacteria is in each of these places, just what kinds.

    In order for this to have any validity, it needs to be a sterile petri dish and he shouldn't be breathing all over the agar---there are many things wrong with this demonstration! Use this video only if you want students to point out what he did incorrectly--only then is it useful!

    Yeah you could streak the bacteria to isolate individual colonies, but you'd still need a battery of biochemical tests to conclusively identify each one. a gram stain alone just wouldn't cut it.

    Gram staining is most definitely not an easy determinant of what is on the plate. Do you know how many gram-positive streptococcus species (not to mention staphylococcus, bacillus, etc, plus all the gram-negative species) there are? To even have a remotely accurate idea of what is on the plate would require a series of different physiological tests.

    Can someone help me? I have to grow bacteria in my petri dishes for the science fair, and I went to my local science store and got the agar powder and I made the agar by mixing water with it and then I put it in the petri dish to be hardened. I swabbed all my surfaces and put them in there. It has been three days since I did that and I'm not getting anything. All I have is like a clear colony in 2 petri dishes, noting colorful or exciting like what he got. Someone tell me what I did wrong? Thx!

    Do you need to culture the petri dishes in a heated area or will they develop naturally at room temperature? Would it be better in a 100 degree environment?

    if you need help building those let me know. I can teach you how to extract Uranium from pitchblende using Nitric acid, I can teach you how to build the centrifuges used to enrich the Uranium. I can also teach you how to mix uranium with Hydrogen flouride to make Uranium Hexaflouride. I can also teach you how to make rocket fuel using ammonia and bleach just in case you needed a delivery system.
