Could you just clarify me in 27:36. When you say n-terminus and c-terminus in the MCS, how could that be possible? Don't they only exist in protein chains? How can the tags be put in n-terminus or c-terminus? Do you put them before translation? But then how do they bind to the resultant protein? So these proteases are a post-translational treatment, how do you insert them into the host? Plasmids?
