Very good presentation, but if the viral antigen can be detected by elisa, why amplification has to be introduced? I want to know in which case this specific technique is used?
IC-PCR is more sensitive than ELISA. It targets the virus itself, specifically its nucleic acid (usually RNA). It can detect very small amounts of viral nucleic acid (due to amplification), making it a good choice for diagnosing infections with low viral loads. While ELISA offers good sensitivity, but may not be as sensitive as IC-PCR for detecting early infections. That is because ELISA cannot distinguish between past and early infection and IC-PCR can detect early infection.
@@kalpna_yadav how does it detect early and late antigen with amplification, can you explain? And I asked about the practical usage and application or kits available? If you know any kits would you state any?
PCR amplifies the target RNA exponentially with each cycle. Ct value indicates the cycle at which the amplified RNA becomes detectable. A lower Ct value signifies a higher starting amount of viral RNA. In early infection, when viral load is high, Ct values tend to be lower. Conversely, higher Ct values suggest a lower viral load, potentially indicating a later stage of infection when the body's immune response might have started controlling the virus. Application of this technique is already mentioned in the video. Kindly watch complete video. And yes the kit is available. You may check ACDcap Immunocapture Kit. The ACDcap Immunocapture Kit is available for use with the following viruses: Arabis Mosaic Virus (ArMV), Grapevine Fleck Virus (GFkV), Apple Mosaic Virus (ApMV), and Cauliflower Mosaic Virus (CaMV).
@@kalpna_yadav thanks kalpana I watched the applications part of the presentation, I just wanted to know the kits available in market, and thanks for explaining the amplification based detection Immunocapture ELISA (IC-ELISA) and polymerase chain reaction (PCR) are both diagnostic tests, but they detect different things: IC-ELISA: Detects pathogen-specific antibodies PCR: Detects the pathogen itself by detecting its RNA or DNA According to a 2000 study, RT-PCR has higher sensitivity and positive diagnostic value than IC-ELISA, but IC-ELISA has higher specificity and negative diagnostic value. However, PCR-based methods have limited application because they lack specificity for the infectious sporozoite stage. PCR can't detect foods that don't contain DNA, like milk, egg whites, and oils. ELISA or LFD tests are better options for detecting allergens in these foods. PCR also can't answer quantitative questions, and only reports the presence or absence of an allergen. I hope it clarifies what I was trying to say, the explaination you gave was about human immunology and the kits are available for plants only, kindly read more about the same.

MP is not doing anything not recruiting forensic students for any post even for a small lab assistant post you need 2 year experience. (From govt organization) & it going in an endless loop they don't give jobs without experience & won't be getting experience without a job) & how we'll get experience?? 2nd thing other South states they recruit on the post of scientific assistant without experience but local language is mandatory.. So basically you will not be eligible anywhere
