It's really a pivotal scene. As the architect of the scam, Billy is taking a huge risk by implying that the relationship with Claire is a hustle. If Virgil was to believe him and walk away, Billy would have lost all the time and money he put into the con. BUT.... Billy knows that Virgil does not respect him or what he thinks, and will ultimately ignore him. So the whole scene with Billy serves as an additional slap in the face to Virgil: Billy's like, I hustled you out of all your best shit and I even told your dumb ass it was a hustle while I was doing it.
This is a pretty horrifying scene knowing the movie's ending.
