Preparation of ICSI dishes
ICSI: injection of the spermatozoon into the oocyte
Oocyte handling (using fresh mouse oocytes)
Learn how to immobilize the sperm and how to deal with oocytes (putting the polar body in the right position to avoid spindle injury) and finally how to perform the sperm injection (practical). Candidates will be provided with plenty of hamster/mouse oocytes and injection pipettes to make sure that they become proficient with microinjection technique.
Dish preparation for ICSI.
Sperm preparation by gradient/ swim-up method.
Alignment of micromanipulator & loading of Micro tools.
Oil loading into the injectors, Equilibration of micro tools.
Loading of sperms and oocytes on to the prepared ICSI dish.
Sperm Immobilization & loading of immobilized sperm in the injection micropipette Holding of denuded Oocyte and Performing ICSI.
