The longevity sirtuin - what you need to know about SIRT6

    Sirtuins are highly conserved proteins that are involved in a variety of important cellular processes such as DNA repair, metabolism and circadian rhythms. The mammalian sirtuins (SIRT1-7) are a family of proteins that carry out NAD+-dependent protein deacylation and mono-ADP-ribosylation. These modifications on proteins can influence their stability, localisation within a cell and activity.
    In the late 90s interest in sirtuins bloomed as it was found yeast lived 30% longer when they had an additional copy of a yeast sirtuin, Sir2. Similar studies have now been performed in mice, but whilst overexpression of SIRT1 in mice does not result in lifespan extension, overexpression of SIRT6 does. This has led to SIRT6 being referred to as the longevity sirtuin. However, there seems to be some sex- and mouse strain-dependent differences. So, in the remainder of the video, we will discuss what you need to know about SIRT6 including it's proposed cellular activities, it's association with longevity and how SIRT6 activation using allosteric activators could have future therapeutic potential.

      I find it interesting that men benefit more than women. If true in humans it might just equal everything out being men on average die before women. Therefore might make it to where a lifelong couple might have a better chance at leaving the world together after being together even longer. Kind of poetic.

    Awesome video as always! I’m currently taking fucoidan, quercetin, and fisetin daily because I think there’s good evidence these molecules extend healthy human lifespan at proper doses and it turns out there’s pretty good evidence that all 3 may be SIRT6 activators which would partially explain why they can extend healthy human lifespan.

      Fucoidan does so many interesting things. I take it for anti-autoimmunity and NLRP3 inhibition - really helps with the 'aches and pains'. But studying it can be problematic as it varies in size and composition from different sources and seaweed extracts can also contain stuff like dieckol and fucoxanthin.

      Most SIRT6 activators, fucoidan included, are at "in vitro" stage. Is there any in vivo or clinical trial SIRT6 activator study?

    You can activate sirtuin 6 with cyanidin which is found in red cabbage and cherries.

    Have you thought about looking at bloodresult variations and aging? E.g markers of inflammation, blood urea, WBC, RBC etc and their changes over time. How they indicate internal aging and what we can do about it?

    Question: Being from mixed parents...could one parents genes be latent and could NMN bring them out of latency? I'm asking because of incredible results I'm getting nfrom NMN incuding white hair regaining colour which starts off the same colour as my mums hair, hazel, where mine was always really dark brown.

    I have a question about NAD+. People get infusions of NAD+ as boutique designer medical service, but even if you did that weekly, is that NAD+ reaching and influencing your cells? Or is that infusion just wasted cash?
    Similarly, does taking MNM or NAD+ benefit a person and work as hoped for anti-aging?
    I’m dubious but also hopeful and curious.

    Since DR (or CR) are said to function by activating Sirtuin genes, does that mean that female test subjects respond less to DR than males?
    Also, at the present time, is it believed that resveratrol has any health benefits at all?

    my mum has been taken the donotage SRIT6 and she got extremely ‏exhausted, as a result was not able to work and not able to cope with the daily routine ,,,,i advised her to stop and see what happens and guess what she gradually recovered and back to normal routine ,,,,,NOW i don't know a thing about science but i know what it did to my mum..... if you ask me this is some dangerous staff

    I have wondered about something for a long time - - butyrate is a general HDAC inhibitor; sirtuins are HDACs. What's up with that?

      As far as i was aware, butyric acid is more a class I HDAC inhibitor, with sirtuins being class III. But on from your point, sirtuin inhibitors are being researched for treating other diseases, so yeah, i think it is still unclear what is going on and likely tissue/disease-specifc

    I would suggest giving more credit to the lab groups, authors, institutions, and journals. These studies are often progressions of earlier studies by the same labs and principal researchers and not one off studies. It's not that hard to give credit where it's due.
