If I were to upgrade the internals of my microscopes led to handle 12 volts would the original 5 volt female jack work? Or are jack inputs interchangeable so as long as the polarity (center negative) is the same.
Según el vídeo, parece ser que el pin en L es el positivo y el que está más centrado es el negativo. El tercer pin parece que no hace falta, ¿qué uso tendrá?

Yes, the other two are negatives but when you insert the male jack the connection between these two negative terminal will break.
