      I’m trying to extract saponins from plant and I was considering using an alcohol like methanol or ethanol as the solvent while I use maceration process for the extraction

    2:43 please.. have a question.. what type of solvent you have used for maceration ?
    I have another question : for ex if we use ethanol-water as solvent, after the extraction we eleminate the ethanol first using ratary-evaporator,but how we eliminate water fraction ?

    Hello po! Thank you po for the informative video! It really helped us understand the methodology needed to conduct our research. May we ask po how much plant material (in grams) do we need to use to extract 5mg, 16.25mg, 35mg of the pure extract? And also how much solvent is required for maceration?
    We are currently struggling to compute the amount of grams of plant material to get the according milligrams (We tried using the vague references in the video but still struggled). Also po we need to compute the amount of solvent used for maceration and the ratio is 1:2 (w/v). The volume of our final solution is 50ml and we need to dilute it for 3 concentrations (10%, 32.5%, 70%). After solving this, the grams needed for the dilution is already stated above (5mg, 16.25mg, 35mg).
    To conclude, our group wants to know how much grams of plant material is needed for the final product using the 1:2 ratio for maceration and how much solvent is required. **We know that this request is a bit hard, if it cannot be answered, do you have any suggestions po on how we can compute it?** Thanks po doc!!

      By “fortify” do you mean purify? You need to do chromatic profiling, separation and purification techniques for that.

    Hello! Thank you for this informative video. I have 2 questions to which I hope to have an answer to, it would be very much appreciated.
    1- In our lab, we don't have a freeze dryer (lyophilisateur), and in a lot of references, i read that they used a drying oven at 40°C for 48h to evaporate to water in the extract. It is okay? Will it not affect my results too much?
    2- For your alcohol extract, after using the rotavapor, the extract was liquid meaning there was still a little bit of solvent in it no? if i'm right, how will you then calculate the concentration if you don't know the initial concentration of your extract?

      1. Yes, you can do that.
      2. Yea, there will be some alcohol left. But you need to further evaporate this in a fumehood for a couple of days.
      Concentration of what? For what? It is automatic that the concentration of the extract you have collected is 100% pure crude extract. If you want to use it as a treatment for testing, then compute the specific concentration you need for the testing using weight of the extract/volume of solvent.

    This video is very insightful. May i ask if you don’t mind, when using the decoction method what is the reasoning behind freezing? And if necessary how will it be possible to obtain a liquid form of the extract instead of the powder? Thank you for any response and for this video.

      Hello! Thank you for your comment. To answer your questions, 1st. the reason behind freeze drying is to remove the water content from your extract without deteriorating your bioactive compounds. 2nd. if you want to obtain a liquid form of your extract, it's either you can dissolve the powder in your treatment vehicle (solvent used treatment, depending on your experiment), or you can use the extract automatically after decoction and filtration, but you have to make it more concentrated. If you don't concentrate your extract, it will probably yield lesser bioactivity.

    I have a question, the powder that i have used for the maceration can i still reuse it? After filtering my solution, there was still leftover solute. After some time, it became dry again. And so i was hoping if i could use it again to make another batch of the maceration process. My plant material is very limited, you see. I used methanol as my solvent btw

      You can but the amount of bioactive compounds might be lesser than the first batch.

      1 and 2. You can only use water in decoction method. You’ll be extracting flavonoids and phenolic compounds that are soluble in water only.

    I'm a 4th year pharmacy student and want to be involved with pharmaceutical science research. I've been thinking about taking a Masters in Pharmacy, but I'm second guessing because from what I see the type of research produced is usually assays of plant extracts which I kinda don't like. The type of research I want to do is along the lines of polymers, nanotech, etc. (i haven't quite figured it out yet). Do you have any advice for me?

      Hi! You can dissolve your plant extract in distilled water. Then put the dissolved extract in your discs. And include distilled water as a control. Moreover, you can read literatures on how to perform Disc diffusion assay.

    What is the shelf life of the "extracts" when performing extraction process? In my case, I consider Iyphilization/freeze drying the plant to achieve the same texture of green tea before boiling it. I wanted to store a large amount of dried leaves for future purposes and save it for decoction. Will it affect the shelf life?

      You can dissolve those in the flask with your extraction solvent. Put them in an evaporating dish and place them in the fume hood for a couple of days.

      Yes, you can. But first, identify your target anticancer compounds. Next, determine in which solvent they are soluble. Then use that solvent to extract your compounds.

      Actually, mahirap ma-estimate kung gaano kadaming extract ang makakuha mo after extraction. What we do, usually, is prepare as much as we can.. mas maganda na yung sobra kesa kulang. Also, when it comes to the extraction method na gagamitin, nakadepende dapat sa target compounds or family of compounds na gusto mong pag-aral. As I said sa video, different extraction methods will give you different products... and these include volume of tincture/oil/concentrate or yung raw form nung na-extract mo. 😊

      You can use glycerine to extract your compounds from your plant sample if your target compounds are highly soluble in glycerine.

    Brilliant. thank-you. Question.........If you spend $xxxx for that magnificent rotovap shouldn't you have the expectation that there would be no final "air solvent drying" step. (post-roto)? I mean isn't that the one damn job its supposed to do well? Why not just run it longer or spin it faster or add heat to the bath or pull whatever vacuum it takes before you suck your extract into the bump trap?(that last one was humour) This may not be relevant but everything else was very clear. Thanks

      Hmm you’re actually right. But when the extract thicken up it’s very hard to collect from the round flask, most especially if you want to collect most of it (example you have limited resources of plant material). That’s the only reason why we need to save a little solvent. 😊

    Hello sir I have ..I've kept coriander leaves for maceration (for antibacterial property against p.acne) .but only using water I did not use ethanol or methanol with the water ..is it possible to obtain the concentrated extract from it and if so wt should be the temperature maintained in the water bath and also the pressure

    Hi. This is really informative, can I ask what are the exact volume / measurement of the used materials here? What is the ratio of the plant to solvent? Will there be a difference in the result if the amount of extracted plants are bigger? Thank you.

      There’s no need to follow any ratio. As long as you have totally submerged your plant sample in your solvent then you’re good to go. After all, will remove the solvent when you run your mixture in a rotary evaporator.

      @@kaeyaalberich4427 you just have to weigh certain amount of your extract and dissolve it in a certain volume of solvent medium.
      Example:
      10mg of plant extract dissolve in 10mL water will make 1 mg/mL plant extract.

    We are currently working on a research where are going to extract Peroxidase enzyme from papaya to detect glucose in a urine through GOD-POD method, but we are struggling in finding a laboratory that will help us to extract the Peroxidase from Papaya so that we can have a analytical grade peroxidase that we can use for our research.

      Rpm doesn’t really matter. This depends on how fast you want to remove your solvent. Temperature depends on the rotavap’s pressure, and the boiling point of your target bioactive compounds (not the plant extract).

      There is no recommended ratio of solid to solvent when performing maceration because the end step is totally evaporating the solvent. So, as long as you have completely submerged your plant sample in your solvent then it’s good.

    Hello po. Did you used ethanol for your extract or distilled water lang po? How many days did it took for you to get the crude extract po?
    And, do you have any knowledge about how to make the extraction of crude extract from an aqueous solution to become faster?

      In this experiment (nasa video), I used various extraction solvent.
      Crude extract ang tawag sa makukuha mong extract after the extraction. Kung ang gamit mo is ethanol, you’ll call it ethanolic crude extract. If water, you’ll call it aqueous crude extract.
      Maceration takes 3-7 days, depending on your research design. The actual extraction for ethanolic extract takes less than an hour.
      For aqueous extract, you only need to boil them. Filter. Collect the supernatant and freeze it. Once frozen, load it in a freeze dryer. It will take 1-2 days or less depending on your extract.

    If trying to extract Alcaloids say plant has 4 different all have nitrogen but only 1 has an OH group how can a person separate the Alcaloids from each other

      I’m not sure but I think you need to perform steam distillation to get extract d-limonene.. try checking then form of D-limonene and look what steam distillation is. 😊

    Hello! I am currently doing my research in plants. I have extracted it with ethanol extract and already airdried it. However, it contains chlorophyll and I am trying to remove it using a separating funnel with non-polar solvents. But there is a lot of confusion, can you explain how can I remove chlorophyll without destroying my active constituents in ethanolic extract also which non polar solvet is best or if there is any other methods.

    I don't get it, ah why did u have to do the rotary evaporation(?) for plant extraction if the filtered solution on the first part is already the plant extract??

      kinda late, but i think the rotavapor removes the solvent used so that you get a concentrated extract

      Water is more polar than ethanol. If you want to extract bioactive compounds specifically those with antimicrobial properties, better check the literature of what Antimicrobial compounds present in your plant and identify in which solvent are they soluble.

      Freezing your aqueous extract (decoction method) is important to ensure that sublimation rather than melting will occur when freeze drying.

      Preferably dry the fruit to saturate the concentration of anthocyanin. Since anthocyanins are light sensitive make sure to dry it on a dark areas.

      Before introducing you treatment (extract) to animal models you should make sure that all of the extraction solvent (ethanol) has completely evaporated. Usually, you can administer your treatment using the common drug delivery methods like oral, intravenous, intraperitoneal, etc. You can also add your treatment to the feed formulation but it will depend on what kind of feeds you have and its nature (dry/wet). Moreover, adding your treatment to the feed formulation might lead to inaccuracies in the volume or concentration of the drug (extract). So, you have to take note of that too. Like for example, if you want to treat your animal model with 200uL of 10 mg/kg extract, you plan how you can administer this accurately to all of your replicates to avoid biases and such.

    Hi doc! Our study po is the effectiveness of c. ternatea linn in lowering blood sugar of alloxan-induced rats. We’ve chosen po the decoction method to extract the C.ternatea. Can I ask po what specific temp and for how long we will boil the C.ternatea? And, what can we use po to filter the extract after boiling?

      For the temperature, it depends on what bioactive compounds with sugar-lowering properties you want to extract from the plant. But in general as long as it doesn’t rich 100C to prevent the bioactive compounds from degradation would be enough.
      You can use a filter paper afterwards to separate the plant material from the extract.

      Hi! If you have no plan on retrieving your extraction solvent (alcohol), you can just use a typical water bath. Maintain a certain temperature, use the boiling point of your solvent or somewhere close to that temperature. Then continuously stir your extract during the evaporation process.
