Can we add and substract within the dimension values. For eg I want to calculate the difference between Sex Male 50 and Female 30, And Difference would be 20. Is there any formula for that.

That wouldn’t works because then looker studio would show the error “cannot mix metrics and dimension” - hope this explains :)
