From what I remember from reading her diary a few years ago, Anne did actually know about menstruation before she got her first period and was waiting to get it. I believe she even mentioned having vaginal discharge. She talked about how they didn't have pads because of rationing for the war and her mother's tampons were for women who had given birth so it was quite a mess when she began having periods.
