What should i add in the dockerfile if I have a backend folder and the project folder separate and for the project to run, first I need to run the app.js file in backend folder and then later do npm run dev in project folder

there is no directory call 'app' then how it work? and which directory docker uses as working directory?

It will be created when assigned. You can name anything instead of app. Docker will target that directory that you have mentioned WORKING in the command.
