    When you want to release volume into a vial.. you mentioned how you must first go to the first stop than hold ? When you say hold do you mean still have your finger held in place in the first stop ? Then you again until you hit the 2nd stop ?

      Yes: Stop at the first stop. That delivers the full volume you set on the pipette. You will see a tiny amount of residual liquid left inside the pipette tip. To empty the tip completely, move the pipette over a different container then press the plunger all the way down.

    How can we use dial down technique for 100% desired volume

      Dial above the pipette's maximum volume, then dial down from there. For example, on a 200 µL pipette, dial to ~205 µL then dial down to set. The pipette will allow you to do this.

    Can I use universal pipette tips on a pos d pipette?

      No. Pos-D has its own tips, sometimes called "capillaries" and sometimes called "syringe tips." Each Pos-D tip has its own built-in plastic piston. The little metal "jaws" inside a Pos-D grab the top of that inner plastic piston to move it up and down. It's a different mechanism from regular air-displacement pipettes that use universal tips. Pos-D is positive displacement: no air between the sample and the end of the piston inside the Pos-D tip.
