You need to first finish Light of Hope mission. Then a guy will give you a side quest called Life of the Scavengers. I believe this guy is located at the entrance to the bar where that singing girl is located. You need to have the side mission in your mission list to actually have the boss spawn. It will also show the location on the map. The area is located very East of the wastelands
Activate the life of the scavengers mission there will be a point where you take rope and ladder down a big opening. You'll go down for a while and when you reach the bottom the story gives way to the boss fight. Good luck.

I somehow finished that Mission without the Boss showhing up. How is this possible?

It's a side quest. If you don't have the side mission the boss won't spawn.
