MHC class I assembly and presentation

- This short video describes how MHC class I molecules are assembled and delivered on the surface of cells to present petides to T cells. This allows T-cells to determine whether the cell is iinfected with a virus by allowing a T cell to essentially look inside the cell. The immune sytem, T cells and MHC class I molecules play a vital role in protecting our bodies from infection.

The MHC I pathway is also essential to antitumor immunity. Just as the immune system views viral proteins as "non-self", peptides derived from mutated genes are also presented on MHC I and will also be viewed as foreign by T cells.
This is a very well organized and descriptive presentation of a seemingly complex process. It can help all levels of education learning Immunology i.e. undegrad, grad, and medical students. Definitely helped me as a grad student!

Three clearly defined loci are recognized within the HLA complex for classⅠHLA antigens. The principal role of the CD4 molecule is to promote interaction of T cells with class ⅡHLA molecules on antigen-presenting cells. The MHC genes expressed in the thymus play crucial role in the selection of the T cell receptor repertoire during maturation ?

Though I wish to ask why would the peptide loading groove have more affinity to the viral peptide than that of a self-peptide ?
i believe because self peptides do not have the correct conformation and also sometimes the self peptide do not have a chance to be loaded
I think they don't have a higher affinity per se because self peptides are loaded as well. Therefore we have the negative selection of CD8 cells in the thymus so we don't have self-reactive CTLs...

How are viral peptides different than regular peptides? And also the viral peptide has to have a high affinity?
