In a previous con, they described it as a tiny reaction that only Jared would notice because he knows Jensen so well. No one else even reacted on the day. It's probably too small to see (and be funny) on camera.

He was chambering a round and to do that you have to pull the top part of the gun back towards you. There are springs in there so as soon as you let go it snaps back into place fairly quickly. What he's discussing is a time where he didn't have his finger out of the way fast enough and the metal snapping forward pinched and cut him.
