There's an interesting video by "Strange Man" about Pleasance. Linking it to the giant snake found hanging in a tree near by. The following is what I loosely remember from it, I do encourage you to check it out for yourself.
Pleasance is based on a town in Ohio called Helltown, in one of the pictures there's a church and a school bus which is rumored to be haunted. A school bus couldn't appear in RDR2 so it's replaced with a school building as well as the church. There's something to do with a chemical spill that supposedly mutated local animals, including one legend about the Peninsula python, a giant snake. In which it spawned python day, a day celebrated by the locals to this day.
You were correct about the satanists part as the church in helltown has an upside down cross. The chemical spill has been altered in game as a plague.

Maybe this is either a reference to the end of RDR1, possibly another reference to the wild west era, a combination of the two, or something else entirely.

3:28 maybe it means how we kill Arthur by playing the Thomas downed mission where he gets tb therefore he gets sick and eventually dies on the mountain(stfu that’s the canon ending to me) and we replace him with John who dies a couple years later

In Undead Nightmare I’m pretty sure you can wipe out multiple sasquatches before killing the last one or leaving him to cry by a tree,so I think it may have been an idea to that
