    I started, still am, in my garage. I use three 4x8 tents. One is my sterile work space and the others are fruiting rooms. I do all my hot none sterile work in the remaining space in the garage as well as store most of my supplies in there. It works well for me and is producing enough fruits to cover my mortgage throughout most of the year.

    Can you please do an in-depth video about agar isolates. And also different mycelial growth on agar.
    I seem to get random results on my transfers, without finding a pattern.
    A lot of rhizo transfers turn Fluffy, oxygen deprived? 🤔

    I grew a few kits last year and then dove in head first, but I'm pacing myself. I ordered some "tongue depressor" sticks that were inoculated with mycelium and grew those out on really low tek agar plates made from sanitized plastic packaging materials that i had/have an excess of that will fit the sticks.
    I did recently get a trichoderma contam for the first time, but I still have lots of non-contam'd library samples on agar and spawn, which is what i focused on all winter instead of actually fruiting anything. I'm pretty sure I was just not using hydrogen peroxide enough and some subset spawn jars may have been in a cardboard box that probably contained onions at one point. D'oh! lol
    Lesson learned. The last agar transfers I did (some from transfers of the contam'd samples to save them) seem to be clean with better practices and way more H2O2.

    Love this! Although my buckets work well and I have a system to keep them clean (soap followed by wiping with bleach) and it works it is labor intensive. This is great advice. Look forward to more!

      Thanks for watching - glad to hear someone has success with those! I have fruited from burlap bags before which are much more eco-friendly but the yields were so low and it was so labor intensive that it was not worth it. I imagine the feeling of re-using containers can be just as satisfying because of it’s eco-friendly nature - a labor of love cleaning those! Mushlove!

    I found a source for sawdust and wood chips however I am unsure if it is hardwood or not. Is there a way to tell by looking at the wood/sawdust? When you say you clean all the time with 15% bleach/hydrogen peroxide, do you mean you clean everything that often? is it 15% bleach mixed with hydrogen peroxide or is it 15% bleach to water OR hydrogen peroxide? Trying to gauge how clean I need to be for my grow/incubation areas. I'm local to Denver as well! Thank you.

      I usually do small test batches with new saw dust to tell if the mycelium will take - I clean first with bleach, let it dry and wipe clean with hydrogen peroxide. Avoid mixing these two chemicals - follow guidelines for the cdphe and department of Ag there is a lot of info about FDA guidelines etc.

      We use sawdust that's a mixture of hardwood and softwood and it works great. You however need to supplement the sawdust to maximize your yields

    So to maintain healthy genetics over time, I would simply take a tissue sample from really desired fruits and place them on agars? Would spore prints be necessary to take so I get a good mix? I think of this as seed saving. I know, for example, to keep pumpkins genetically clean, you are to save seeds, not only from one pumpkin, but dozens, or 100,s, I am not sure of the exact number, but there is a number with every different type of fruit. Any of your knowledge appreciated😃🌈🤙

      Yes spores are like “seeds” and can create newer stronger genetics and tissue is a clone so will produce consistent results but will slowly degrade over time

    cant you use a UV light to sterilize? Also it would be way more helpful if, instead of generalizing, you would actually mentioned which brands or companies should have better genetics.

      you can sterilize surfaces with uV but it will not penetrate substrates. I don’t know every brand out there - I can recommend fungi perfecti, north spore, mycelium emporium, amacel, and some other youtubers out there.

    10:00 Where did you get the idea that mushrooms senesce? I know stamets mentions it but others like Peter McCoy in "radical Mycology" basically says that mycelium doesn't senesce(at least common gourmet mushrooms) and the loss of vigor can be attributed to enzyme deficiencies among other things and not senescence. If senescence was a thing huge clonal colonies of certain mushrooms could not exist, hell certains old strains in the mushroom cultivation world wouldn't be possible either. I have read good arguments for both but there is really no clear cut answer as far as i can tell. Do you have any good info on the topic of senescence that i could read?

      Generally, the cells genetic structures (DNA) will break down over time due to natural imperfections during replication of cells and the random chance of mutation that is not corrected for by other cellular components. The amount and time that a strain will senesce depends on its own genetics, environment and yes there may be enzyme deficiencies because of the narrow range of nutrients that a culture receives in vitro. But even in the wild this will happen and it is corrected for by breeding (spore dispersal) and natural selection - variances in environment that favor new genetics or vice-versa. There are methods for maintaining viability of cultures for long term that I have not discussed but may do a video in the future👍 Thanks for watching ❤️🍄

      ​@@FreshfromtheFarmFungi I know it's logical to assume eventually every cell will lose the ability to effectively replicate because of DNA degradation and mutacions, this seems to be the norm in unitary organisms(vertebrates,insects,etc) but modular organisms do not necessarily undergo systemic senescence, and do not segregate somatic from germ line cells.
      We know that there are mycelium(for example honey mushroom) that have at the very least negligible senescence because their mycelium network all sharing the same dna can get huge and live for thousands of years not showing any signs of slowing down, and there are many other examples in plants like "pando" and many other organisms like algae.
      There are fungi that do show clear signs of senescence being loss of vigor and ultimately death among other things, but this seems to be an evolved trait where the mycelium expends all it's resources for greater reproductive success, kinda like monocarpic plants, or for a better example some coprophilous fungi that live on a resource that will inevitably run out so there is no selective pressure for it to live past a certain time frame after reproduction.
      Imo common cultivated mushrooms like oysters don't seem to experience senescence, at least not to a degree and in a timeframe that would affect the mushroom cultivator.
      In my experience i have been expanding the same spawn of oyster for close to 2 years unrefrigerated and have yet to notice any loss of vigor in the particular strain, so for me it's hard to believe that a loss of vigor after 20 or even 50 agar transfers can be attributed to senescence or ageing.
      If you think about it any strain you have probably came from a guy who cloned it from a wild specimen who sent it to a lab that grew it out and when to another lab that did the same thing ... etc... to finally a guy that makes spawn that has been using the same slant for years that sold you your spawn. In all that process i can't imagine anything less than 20 agrar transfers for that mycelium in reality probably way more.
      So i don't think commonly cultivated mushrooms eventually die though i am open opposing evidence, but i do think that conditions like enzyme deficiency could be real and maybe even the reason people think mushrooms senesce.
      May i ask where did you come upon this idea specifically for MUSHROOMS? Could it be one of Paul Stamets books? Because i think in one part he talks about some lions mane showing signs of losing vigor in as little as 8 transfers, if senescence was to blame we wouldn't have 20 year old lion's mane strains that still produce great flushes no matter how well preserved they are kept(cooled liquid nitrogen or whatnot).

    Can you use coffee grounds as substrate?
    Also, I've started growing but I simply cannot get my shrooms to pin. I just purchased a humidity gage and a mini tent for humidity but they have not arrived yet. I have had these in my bedroom with the ac on and at first, I think they dried out because I had the lids slightly off. Now I'm not sure what's wrong as the humidity is fine, but im not sure about fae. I cant figure out how to strike a balance and get these guys to pin, I think I may just toss them. I only got one small side pin, but im not sure how to get a good environment. Any suggestions? I live in the midwest with only one ac, its generally around 80 degrees inside unless i have the ac on...
    Sorry for all the questions, its been super frustrating.

      Thanks for following! I have used ozone in the past with plant tissue culture and was impressed by it’s efficacy. However, some of the plants suffered with developing lesions and spots on the tissue so we stopped using it. I have not explored ozone with mushroom cultivation but my feelings are that it is indiscriminate and can hurt whatever is in the room including people and plants/fungi. It might be effective between batches to clean grow rooms etc. but would not recommend regular usage during cultivation - just my opinion I have not tested since the initial experimentation about 6 yrs ago.

    Dose anyone have advice on what to do is your mushrooms are in a shoebox type box and growing pins on the sides and bottoms of the box? Should I flip it? Move it to a bigger box? I’m confused on if I should do anything or not but there’s about 20 on the sides that might not make it to the top. One pin already has, it’s kinda mushed up between the side of the box, that’s the only one that’s made it past the substrate though. I see them on the sides and bottom, there’s a little bit of space between the edges and that’s where the pins are thriving but they’re growing downward and sideways not upwards.

      next time put a liner in there, as the substrate shrinks it creates a micro climate that produces pins on the sides but if you have a liner it shrinks with the substrate and prevents this - happy growing!

      if it grows out on grains they will become deformed but they may have a chance for bulk still if they are just pinning

    Hey man, thanks for the nice content. I was wondering since one of your points is ' dont cut back on sterile techinique', What is your opinion on the low tech farming grocycle is teaching. Can this work or do you think this is a no go for starting off? Grtz

      In other words, if properly managed, can a farm work without sterile technique? Like choosing the right varities that colonize fast, in order to get a cashflow without large investment... Lime pateurisation is one of the techniques they use i guess, in combination with fast colonizing oysters

      it can work yes but you will soon see how inefficient it is - with low tech growing methods you are trading energy, man hours and lots of contaminated substrate for up front investment to do it more efficiently with sterile techniques. It might be reasonable to learn this way and do it small scale, then once you get your feet wet upgrade where it pays off most. It happens in nature all the time so I don’t want to knock it but there are better and more profitable ways - Good Luck!

    Hi could you help me, im trying to find a video showing me how to remove mycellium when you have too much of it,and is it possible to move it to another medium to grow it instead of wasting it. I bought a grow at home kit, i live in the UK and for once we had a slight heatwave which has sped up its growth...a little too much. Its my first time.

      probably doing it too fast or too crowded or maybe they are contaminated with yeast - try washing them lightly, patting dry and spacing them out more

    Does multispore LC compete with each other while growing in grain/ substrate. Am I getting 1000's of genetics near the end...or just a dozen that never pin lol....

      it may be that one out competes the rest and is finicky or sensitive to environmental factors. There can be many mutations in LC spore solutions so it’s hard to say for sure. Best to repeat many times to avoid this

      I run my furnace fan constantly when doing lab work just make sure you have clean duct work and change out filters regularly

      as for fruiting, Try to make the whole air exchange and environment compartmentalized and separate from the rest of the inside - exaust outside for sure

      I am at 6000 ft. This is one of my earlier videos just doing one take and learning how to do them - the info is there and it’s solid advice. I think breathing in between thoughts is important hah

      @@FreshfromtheFarmFungi Lol, 6000 feet is up there for someone like me who lives by the ocean. I understand what you mean because sometimes I get the same way and I get so excited I forget to breath when trying to get me thoughts out.
      I remember being in the mountains in Wyoming and it was hard to catch my breath just standing still, then I tried to ski. 😂

      you should shake them in solution to separate or use a streak method to separate the spores

      Clumping in a syringe is normal. I bet your spores are fine. Knock up some sterile agar or jars and you will know.
