They are not both correct because the letter N (ん) exists in Japanese alphabet, while the M sound alone does not exist: if you try typing M on your Japanese keyboard, it won't write any sign:
せんぱい (with the N)
せmぱい (with the M, which doesn't exist as a single letter: Japanese alphabet is made of syllables.)
So seMpai isn't correct and it doesn't mean "lover" either, because it actually doesn't exist as a word because M isn't a syllable, while N is considered as one of them.
