Epigenetic Clocks Help to Find Anti-Aging Treatments | Steve Horvath | TEDxBerkeley

  • Steve Horvath is a professor of human genetics and biostatistics at the University of California, Los Angeles. Horvath had a lifelong interest in solving an important problem in aging research: how do we measure aging? In 2011 Horvath and his collaborators at UCLA described the first age estimation method (epigenetic clock) for saliva based on chemical modifications of the DNA molecule known as DNA methylation. Two years later Horvath published an age estimator that applies to all tissues and cells of the human body. This discovery, known as the Horvath epigenetic clock, was unexpected because cells differ greatly in terms of their epigenetic patterns. Recently, he has studied treatments that slow or even reverse aging in humans. He and his colleagues have demonstrated that the epigenetic clock predicts lifespan and is related to centenarian status, obesity, HIV infection, early menopause, progeria, and many other age related conditions.     Steve is modest as ever but the universal pan tissue epigenetic clock ought to be called The Horvath Clock in his honour. Wouldn't surprise me if he wins a Nobel Prize some day.

    I like that Steve and his friends, in high school, decided they wanted to work on stopping ageing. My friends and I just wanted to graduate without getting kicked out!!

      @@joshtuk Liz Parrish is a name worth mentioning, also David Sinclair and George Church are prominent geneticists in the field. These are the people who I'd say are probably working hardest to realize the vision Horvath laid out.
      There's a growing body of research papers in the field, Dena Dubal is probably the most prominent who doesn't have much of a media presence, if you have the patience (and who could blame you if you don't) for complex research papers, hers are very well written.
      And if you want, I'd be glad to talk about this with you in more depth elsewhere, I don't know if you have Discord or what have you, but I'll try to add you where ever.

      @@joshtuk A couple names worth mentioning besides this guy, Liz Parrish and David Sinclair are both independently looking for a cure to aging, and have both made significant progress. There's a lot of material on this and a lot of voices, those are the ones besides Horvath who have stood out the most to me.

    Interesting research. Would be fascinating to see a twin study of methylation. The fact that we can slow aging with behavioral change is hopeful. Even if it means my wife is right about the veggies.

      George Church’s company Rejuvenate Bio is developing a gene therapy to reverse aging (or aspects of aging). They have very impressive results in mice, and trials in dogs are already underway.

    This is a great discovery and Steve seems like an awesome guy. But I wonder if DNA methylation is accurate in anti-aging treatments? People have reversed their epigenetic age by taking molecules such as NMN, meaning that their DNA methylation levels have gone down, but does it actually mean that physically the same they were as before? In other words, if I am able to reverse my epigenetic age by 10 years, I should look and feel exactly as I was 10 years back. But from what I have seen, it doesn't exactly work like that. So I guess it's possible to trick your body into having lower DNA methylation levels without actually reversing the physical state of your body in entirety.

      I don't think you can ever go back to a younger state, but you can change your cellular and molecular age so that your cells don't age and die as early as they would have without the treatment. So you will live longer and post-treatment, you will not feel as old as you should have felt

      In Greg Fahy's TRIIM study (briefly mentioned in Steve Horvath's TEDx talk), 3 out of the 9 participants actually regained color in their hair after 12 months of treatment with growth hormone, DHEA, and metformin.

      Yes, I believe yoy can feel 10 or 20 years younger with some supplements and a positive outlook on life. I definitely feel more than 20 years younger now. Last 10 years, I have gone through a cancer operation, broken right hip, took a long time for an operation date, so left hip deteriorated, which again I had to wait 5 years. After my left hip operation, I have been getting stronger with weight training and gained flexibility through daily strethching, pilates and yoga practice. The right supplements have helped me with my workouts, gradually getting better than 20 years ago. NMN, Rervatrol, TMG, vit d, k2, b12, creatinine, ALA, magnesium. I wish to remove hypothyroid problem within a year and osteoporosis on my next dexa test. I am in my 60s. People do not believe my age. I have no lines or wrinkles, but a faint line going from either side of my nose, which I hope will go away gradually. NMN, reservatril and Tmg only taken for about 3 to 6 months on and off. Cheaper versions of them.

      In UK, we cannot get metformin. I take berberine and I believe that has helped. Also omega 3 supplements. Apart from last 2 weeks Christmas weeks, I do not go for desserts and sweets. Well today is 31st Dec 2022. From next week back to my normal diet, mainly veggies, salads, nuts, fish.

    I still don't understand the point of epigenetic clocks. It'd be one thing if the were designed to predict at what age we are likely to die, but I really don't see the point of learning one's current "biological age." For instance, I'm 36. Let's say an epigenetic clock informs me that I'm biologically 39. So what? Am I going to live to 85 or 110? What does being "39" when I'm 36 actually tell me? Nothing useful as far as I can tell. At most it might imply that I'm 3 years behind the trajectory of the average person. So does that mean I'll die 3 years later than average? Of course not. So what does it mean? Anything?

      I think the point of an epigenetic clock is - to be used as a reliable tool (instrument) that measures the efficacy of different medical interventions aimed at age reversal.
      I don't think it was ever designed to predict one's absolute age at death.

      @@cosmotagliafierro8055 Whatever the original intention, it seems they've been somewhat hijacked by biohackers and longevity researchers while they're still largely in beta, and well before they've reached "useful" status. In any case, if
      they're not designed to predict the probable length of one's life, it seems difficult to determine some other useful frame of reference that would make them meaningful. Without using my date of death as a point of reference, I have no idea what being "biologically 40" means. It's like being told your time during a race without knowing how long the race is.

      @@cosmotagliafierro8055 why do we need to pay for an epigenic clock. From all the biomarkers at my doctors, the nurse told me by biological age. Also a lot of gyms have machines, which scans your body with various biomarkers and tells you your biological age. It looks at muscle mass, fats, strength and tells the age. These 2 are ok for me.

      Accent is a product of the age of a person when they learn the foreign language. If you learn at say 7 or younger, there will very little if any accent. If you learn at 15 or later, it will be far more noticeable. Though there are certainly exceptions. Being a perfectionist, a terrific student and having excellent hearing, probably helps.
