honestly mj was the smartest “girlfriend” or female friend from all the spider-man movies. like mary jane and gwen never figured it out and mj just used her brain and connected the dots.

It was important, because he reacted in two totally different ways: one was really not true, one he was trying to hide. To the think you know is definitelly not true someone reacts in some way, then he reacts in another way to the think he wants to hide, wich leads to conclusion. That is what that scene means.
