Some add more damage on Servants weak to Enuma Elish (one Buster) by 30%. Another Buster gives him a healing boost for 2 turns, the Quick raises C star drops, the third Buster adds 1000 of flat damage to a hit and the Arts automatically drops 4 stars. You have to use the cards for the effects to work.

It's possible, but even with a ton of debuff, the chance is still really small *I insta kill Gil once, but it's only happens once in my 2 years experience
If you have EX+++++++++++++++ rank luck you might even get an one shot with Hassan since he has a passive chance to inflict death that a terribly small chance of working. Got it once during Babylonia against Quetzalcoat

Solomon, King Hassan or any of the Grand we don't really know who are all the grands and who is the strongest between them. As for normal servants Serious Gilgamesh is the most likely one with Enkidu, Karna and Arjuna not being far.
Kiara is just like a ton of other Servants they will fight you in différents events but will be your ally if summon if Chaldea. This version of Kiara is her trying to take the universe so in this case she is 100% enemy

Kiara is a special battle in which you can seal her abilities by buying stuff in the shop, which really helps to make the fight managable.

For gameplay reasons, Instant Death is very low against bosses. If you get it your luck is more than a 5* Servant roll
