No not advisable to use diverter on second floor. You will need a pressure pump .

What will be the height difference between bottom of the overhead tank and the inlet of the diverter. It has to be 30 feet or more .Or you can use a High Flow Diverter from Jaquar brand. Strongly recommend wall mixer and not diverter .
Also
Install a sand filter in the main line taking water to the overhead tank and a magnetic water conditioner.

No. Nothing happen like this.
Yes it can happen if there is a pressure differential. What kind of diverter are you using or plan to use . In single lever diverter the chance is very low but if you are using 4 way diverter then you have all the chance of this happening .
Single lever diverter needs more pressure
Four way diverter will work with low pressure. Jaquar has a 4 way diverter which has non return valves to stop cross flow of water .
Wall mixer has 100% chance of cross flow . You can install non return valve with wall mixer to prevent this .

We instal diverter required pressure pump or not required
To give you a proper answer we need to know more details of the height of the overhead tank and number of bathrooms serviced by this tank .
If you want a hotel like experience then you need a pressure pump.
If you have already done this then install
1) Pressure Pump
2) Sand filter in the main line taking water upstairs to the main tank .
3) A magnetic water conditioner to tackle scaling and hard water .
