How does he play comfortably/improv in an alternate tuning? Doesn't it take a lot of time to get used to finding all the notes and everything?

If anybody has the banjo tuning for Mystery of Love, then I can die a happy man.
Possibly a bit late, but only just saw this comment.
Without the capo it's Bflat DGCD. Then capo on 6, including the top string.
The top string is 1 and a half steps higher than standard, so be careful when tuning it that high!
You'll have to down-tune it when the capo is on to sound right and play along with this.
Hope that helps.
p.s. I play it GDGCD and then put a capo on 2. Mostly because I'm scared of tuning my top string that high, and I have a lower voice than TTMOE.
