The resort looks huge, is it walkable or do they have trolleys to shuttle you around ?

Yes, the resort is huge. I have been to so many resorts and I am not sure if there is a trolley at Grand Sirenis, but some walking may be required between the rooms and the beach and pool areas. Shuttles may exist for other areas.

If the restaurant is full and you can't reserve for that day, where are you supposed to eat?

There's always space in the main cafeteria.

I do not know. I live in DR so I have not had to travel for a bit. Outside of the resort, it is normally between 1300 and 1800 pesos and it can be up to 2500 pesos if they come to your house or hotel. Some hotels in Santo Domingo offer the service for free.
