Please explain where the excitation source, UV and fluorescence and filters are during a measurement? It appears that a flash of light goes from well to well from the top of the plate; the UV detectors are on the bottom and there is some type of pivoted mirror that directs the fluorescent emission through a filter to the detector.

One small question: let's say you connect the part just upstream to the ORF (composite promoter, 5' utr etc to the ORF. Then in order to have a universal overhang, you need to introduce a few "foreign" bases before the tag, right? That may affect expression levels....
