in TEM , magnetic lenses are used instead of glass lenses in light microscopy. IN EM, you can not see actual specimen , what you see is image of specimen. EM operates under vacuum to prevent electrons defections, in EM, source of illumination is electrons of narrow wave length, whereas in light microscopy, visible light used. this is the reason for higher magnification in EM. in EM, the magnification is enhanced by increasing voltage .these are few major differences. In EM, negative staining is used due to which you can get shadow of specimen in black and white, never in color. in EM, carbon coated copper or polymer 300 mesh grids are used instead of glass slide in LM. these are few important differences
