    If I may add something to the discussion at the end: - Normal HPLC systems using metal capillaries etc. can be used in combination as said. This usually works fine. If problems with noise should occur, passivation with acid, as said, helps, as well as using PEEK capillaries between the HPLC and ECD. But as said, it's usually not a real problem. The same is true for columns, metal columns can be used. A fully metal free system is really only needed for trace analysis capable of measuring tiny concentrations, but in those cases you'll go for an ideal system reserved for that purpose system anyway. - Sensitivity difference between ELSD and ECD is a very good question and I can't answer it too^^ The ELSD is rather unselective, if it's not volatile, it will form a mist that scatters light, which is very dependent on the specific molecule, so not universal at all, but still all analytes will be in the same ballpark. The ECD on the other hand is pretty much the opposite, the detector response is very dependent on the molecule itself (especially the functional groups) and the method parameters used. Altering them can get you anywhere concerning response and noise. So there's really no answer to this question. It's really specific to each analysis. The ECD can be better or worse depending on analyte/matrix/method. An expert (Freaky scientist developing methods on it since multiple centuries type) may be able to evaluate the more sensitive detector without experiments, but us normal humans, we try by experiments if we have an ECD or else ask Google 😅 You can make good assumptions but there are just to many variables with big contribution to the overall chemistry, so if there's no "won't work at all" on any side of detector choice, you'll have to find out experimentally. To be honest, I try to avoid the ECD because it is finicky, but there are cases you have to go with it. Either regulations (pharmacooeia) or either its selectivity (ECD vs. an expensive purification step in the lab) or sensitivity (it is a very sensitive detection method, but very small seeming factors can add incredible amounts of noise).

    17:41 I don't know the method used, but I'm pretty sure back pressure isn't a factor here 😊 ion or ligand exchange chromatography is mostly done at very low back pressure due to the separation mode and material used for it. The high temperature in sugar analysis is done because of the tendency of sugars to form an equilibrium of different anomers (same molecule, same isomer, but different ring geometry). The temperature pushes the equilibrium to one of the two forms, because they usually elute differently and therefore form two peaks instead of a single one 😊 Edit: They still are very sensitive to pressure though. Those columns should only be used with slow flow gradients and the temperature change should only be done with a small flow of eluent, because the volume contraction and expansion can damage them. So make sure to only heat or cool them while pumping a small flow of eluent through them. So it's not completely wrong. Edit 2: The gradient from water to NaOH solution is very interesting. I've never seen such an application but it totally makes sense 😀

    Interesting video, thanks for posting it! Before seeing it, I've never heard of Knauer. I'd guess Antec builds your ECDs? Based on the similar look and them being near Germany and leading in ECD. ECDs are pretty amazing detectors, especially concerning their selectivity and sensitivity, but also very finicky and hard to use. Amazing for experts in the field of electrochemical detection, but sadly a pain in the... in routine analysis, especially if you need a gold electrode instead of the glassy carbon one. A great detector but in need of a lot of knowledge and the necessary carefulness. Edit: The RID/ELSD/(CAD)/ECD comparison is very true. The RID is rather easy to use, isocratic and simply works. But lacks sensitivity. The ECD is very difficult to use but offers way better sensitivity. The ELSD as well as the CAD are very different but often used in similar applications where there is no chromophore für UV/VIS detection, can be used with gradients and they're both pretty much between the RID and ECD concerning the ease of use and their sensitivity.

    If I print directly, without a PC , where do I assign sample name? is this instrument part 11 Complient?

      Without a PC it isn't possible to enter a sample name. Therefore, there is also no information on the printout. A sample name can only be entered in the Software, which unfortunately is not CFR 21 part 11 compliant.

      As raw materials like mRNA and lipids are very expensive, most customers want to start working with very small volumes, especially in R&D. For these small volumes it is very versatile that the NanoScaler is flexible regarding the used flow rate.

      Ich denke dadurch wird der zumeist bescheidene Chemieunterricht interessanter. Ich hab mich damals nur für das Chemiestudium entschieden wegen dem was wir in der 10. Klasse hatten. Ansonsten fand ich Chemie recht lästig ^^
