Quick question. Where are your gloves?
Great question...years ago no microbiologists wore gloves! In fact, dentists didn't wear gloves until the '80s. Even chefs that handled food didn't where gloves. We also mouth pipetted bacterial cultures. How things have changed!

What is the recipe for transfer buffer?
We now use Bio-Rad Transblot Turbo, which only takes 7 min or 10 min or 15 min depending on the protocol. Here we were using 12mM Tris, 96mM Glycine, 20% Methanol, pH 8.0.

These do appear someone brownish in my video camera, but both of these strains are alpha-hemolytic and have a dark greenish tinge. I'm sorry it doesn't look better.

The one not susceptible to the bacitracin could be group B or C streptococcus. To test group B use SXT (sulfmethoxazole trimethoprim) disk. If susceptible, then it is group C. If not susceptible to either A Disk(bacitracin) or SXT disk then you must confirm group b test with a camp factor test.
